Key points of Solid Phase synthesis:-
      The reputable procedure of   peptide synthesis in the laboratory known as solid phase peptide synthesis .

·         Solid Phase Synthesis, quickly linking of a peptide chain through following reaction of amino acids on insoluble solid support by using polymer resins.

·         Polymeric resin beads having some reactive function groups like –OH, NH2.
These are link to amino acids.
·         Amino acid can link to the resin, excess of reagents and amino acid can be throughout by washing and filtration with vacuum between each step.
        
·         Each amino acid couple to second amino acid (it must be protected) if amino acid having side chain, here protecting group strategies have to use.

·         Initially amino acid is couple to the resin ,subsequently amine is deprotected,then couple with another free C-terminus of amino acid .This process is repeats until the desired peptide has been synthesized .this process is  includes washing, capping and deportation.

·         At the end of the synthesis, the crude peptide is cleaved from the resin while simultaneously removing all protecting groups in our peptide.The crude peptide is precipitate from non polar solvents in order to remove unwanted organic soluble by-products
